同型对照(Isotype Control)：使用与一抗相同种属来源、相同亚型、相同剂量和相同的免疫球蛋白及亚型的免疫球蛋白，用于检测由于抗体非特异性结合到组织细胞而产生的背景染色。
如果一抗是多抗，可以用an normal serum(与一抗相同的正常血清) (must be the same species as primary antibody). This control is easy to achieve and can be used routinely in immunohistochemical staining.

这才是真正意思上的阴性对照，而国内现状是极少做对照实验，或只用PBS代替一抗进行阴性对照实验。

mouse anti rat CD11b,clone OX-42 的 isotype 是 mouse IgG2a， 所以您要用purified(纯化的) mouse IgG2a来做 OX-42的同型对照(Isotype Control)。
一般的的生物技术公司和国内的代理都有出售。

版主，我现在做大鼠变应性鼻炎模型鼻粘膜中蛋白A的免疫组化，现在国内外没有找到别人报道过，由于经费不够，所以没有没有用ELISA，预实验发现正常组和患病组都有着色（EnVision二步法），问题是：
1、阳性对照和阴性对照都需要做吗？（阳性对照是鉴别假阴性的是吧？既然两组全有表达，是不是阳性对照就不要了？）如果需要，该怎么  设计对照？
2、结果我需要取五个视野，数阳性细胞个数进行统计分析，由于可能在多种细胞中表达，那到时候我是不是要分类计数？听说图像分析系统也可以进行免疫组化结果统计（好像是什么灰度值）
学生初次做免疫组化，很多不是很懂，恳请您的指点，先谢谢您了

Sorry, I get confused here. May I ask: "do u mean use the primary Ab and isotype IgG2a as well to gain a more specific binding?" In this setting, secondary Ab would combine to whatever kinds of mouse IgG and yield to everything positive!

In contradiction, IgG2a could compete with 1st Ab to bind with antigen and only if IgG2a, could be named with 'cold drug', is 100 times concentrated than 1st Ab, named 'hot drug', the bound between 1st Ab & antigen could be blocked to the extend of 99%. Am I on the right track? So this is all about giving a non-specific binding?
